Evaluate retinal health in refreshed ICR mouse colonies.

Retinal degeneration associated with the pde6b mutation historically affected up to 80% of ICR mice, complicating ocular toxicity interpretation. A refreshed U.S. colony—derived from mutation-free stock—was evaluated to confirm elimination of this background lesion.

Genotyping and microscopic evaluation showed that nearly all refreshed-colony animals were wild type, with no retinal degeneration observed. In contrast, discontinued-colony animals showed moderate bilateral retinal atrophy consistent with the pde6b mutation.

This study demonstrates how colony refresh and genetic screening improve model reliability for ocular safety assessment.
