Assessment_of_Factors_Impacting_DuplexSeqLibrary_Performance

Duplex sequencing (DuplexSeq) is an ultra-accurate, error-corrected next generation sequencing (ecNGS) strategy that is used throughout the genetic toxicology community for detection of gene mutations in vitro and in vivo, with a long-term goal of incorporating DuplexSeq (and other ecNGS platforms) into OECD test guidelines. On occasion, a sample fails library preparation or underperforms during sequencing. Our poster, “Assessment of Factors Impacting DuplexSeq Library Performance”, presented at the 2024 Environmental Mutagenesis and Genomics Society meeting, explores the common factors that could potentially affect sequencing performance.

Download a copy of the poster to see the results of our analysis, including:

  • Identification of factors that can impact sequencing performance
  • Relationship between performance metrics and input DNA mass, DNA quality and median insert size
  • Quality of mutation frequency and spectra results from under performing libraries
