The Enhanced Ames Test (EAT) is a modified Ames assay that is used for the mutagenicity assessment of nitrosamine impurities. One of the modifications of the test is to include a mutagenic N-nitrosamine impurity as a positive control in addition to the standard positive controls. Multiple trials with various N-nitrosamine impurities were run to try to establish a standard positive control to include in the EAT. These trials reveal the difficulty in determining a universal N-nitrosamine impurity across all Ames tested strains.
